What PolyPhil™ is.
PolyPhil™ is a polynucleotide skin booster from the Austrian brand Croma-Pharma — a pioneer in minimally invasive aesthetic medicine and active in over 70 countries. PolyPhil™ is manufactured at Mastelli in Sanremo, Italy, the company that co-founded polynucleotide treatment back in the 1950s. The formula is based on highly pure polynucleotides from controlled European freshwater fish (suitable for human consumption) and is delivered via the patented PN-HPT® technology (Polynucleotide High Purification Technology) cleaned.
Polynucleotides are linked chains of nucleotides that occur naturally in the human body in the form of DNA and RNA. When administered intradermally, they act through three mechanisms: Fibroblast stimulation — the skin’s cellular „architects" are activated and produce new collagen and elastin. Antioxidant protection — Polynucleotides bind free radicals and protect the skin from oxidative stress. Hydration — they bind water in the deeper layers of the skin. The result: firmer, more elastic, better-hydrated skin — without adding bulk or altering facial features.
The polynucleotide treatment dates back to Mastelli in Sanremo, Italy – a pharmaceutical company that has been researching DNA-based active ingredients since the 1950s. Vital Injector 2™ with vacuum control — as with NCTF®.
You're lying there, relaxed. The Vital Injector 2™ handpiece gently tightens the skin, whilst the 5-needle system applies PolyPhil™ evenly and precisely. One ampoule (2 ml) deals with either the Eye area, the whole face or Neck and décolletage. You feel a gentle pressure, not a stinging sensation. This is followed by a customised mask, Dermalux™ LED phototherapy and SPF 50.
Ready to go out straight away. No bumps. No downtime.
Unlike traditional polynucleotide mesotherapy, you leave the clinic no wheals, no micropapules, no bruising — a result of the Vital Injector 2™’s precise vacuum control. Make-up can be applied on the same day, but this is not recommended. Avoid exercise, saunas and heat for 24–48 hours, gentle care, SPF 50 every day. The first signs of freshness are visible within days; the full results develop over 4–12 weeks — with improved skin density, firmness and radiance.

What is the difference between PolyPhil™, PolyPhil™ Next and PolyPhil™ Eye? +
All three are polynucleotide skin boosters from Croma-Pharma — each with a different focus:
- PolyPhil™ — 20 mg/ml polynucleotides. The basic formula for pure biostimulation. Ideal for young to mature skin, with a focus on skin quality, skin density, fine lines and acne scars.
- PolyPhil™ Next — 10 mg/ml polynucleotides + 10 mg/ml hyaluronic acid. The combined formula — biostimulation and hydration in one. Ideal for mature or dehydrated skin.
- PolyPhil™ Eye — specially formulated for the delicate eye area. Firms and hydrates without the risk of fillers, ideal for tired-looking eyes and fine lines around the eyes.

How much does PolyPhil™ cost at alestetics®? +
A PolyPhil™ treatment costs CHF 500 per ampoule — including a personalised mask, Dermalux™ LED phototherapy and a relaxing head massage.
One ampoule (2 ml) is sufficient to treat either the Eye area, the whole face or Neck and décolletage. If you combine several areas (e.g. face + neck and décolletage), you will need two ampoules — so CHF 1,000.
In the Pack of 3 you get 3 sessions for CHF 1,200 — designed to fit in with a typical 6–8-week bulking programme.
How many sessions will I need? +
The standard protocol at alestetics® is 3 sessions at intervals of 2–3 weeks as a restorative treatment. Initial improvements in hydration and skin texture are often visible within days, whilst the full results, achieved through collagen regeneration, develop over 4–12 weeks.
We therefore recommend a Maintenance: 1–2 sessions per year, depending on your skin goals and in line with your overall treatment plan.
Is it true that it’s „salmon sperm"? +
No. „Salmon sperm" is a sensationalist term used on TikTok and social media that is scientifically inaccurate. What is actually used is highly purified polynucleotides made from European freshwater fish (suitable for human consumption).
Mastelli's PN-HPT® technology removes Proteins, contaminants and biological residues — what remains are biocompatible active fragments. What is injected are not biological cells, but pure polynucleotide chains — chemically similar to those that naturally occur in your own skin.
The TikTok term is clickbait. What you get is European GMP quality in line with pharmaceutical standards.
Is PolyPhil™ safe for people with a fish allergy? +
Thanks to PN-HPT® technology, all proteins removed — this increases the risk of allergies significantly reduced. All that remains are pure polynucleotide chains whose chemical structure matches that of the body’s own.
Nevertheless, at known fish allergy We assess suitability on a case-by-case basis during a consultation. In cases of severe fish allergy, we advise caution and discuss alternatives such as NCTF® 135 HA, which contains no animal-derived ingredients.
Does the treatment hurt? +
Thanks to the Vacuum control With the Vital Injector 2™, the treatment is virtually painless. Instead of traditional single injections, five needles are inserted simultaneously at a precisely controlled depth — you’ll feel a slight pressure, not a prick.
Most female clients find the session easy to cope with, and many even find it relaxing due to the gentle vibration. For particularly sensitive skin or the eye area, we can apply an anaesthetic cream before the treatment.
What is the difference between this and NCTF®? +
Both are active ingredient treatments administered via the Vital Injector 2™, but with different active ingredient profiles:
- PolyPhil™: Focus on Polynucleotides — Fibroblast stimulation, collagen and elastin production, antioxidant cell protection, tissue repair. Ideal for restorative treatment, acne scars and thin, sun-damaged skin.
- NCTF® 135 HA: Focus on 60 essential active ingredients — Hyaluronic acid, 12 vitamins, 24 amino acids, co-enzymes, minerals, glutathione. Ideal for skin quality, radiance and holistic nourishment of the skin matrix.
We use the AURA 3D skin analysis to determine which treatment is best suited to you. We often alternate between the two within a treatment plan — but never in the same session (with a two-week interval).
Can I combine PolyPhil™ with Vivace® or Volnewmer®? +
Yes — and these are very powerful combinations. The strategy varies depending on the treatment:
- Vivace® RF Microneedling — can be done during the same session: After Vivace®, the microchannels remain open for 4–6 hours — PolyPhil™ can be applied directly afterwards to the open skin matrix. Perfect conditions for the deep penetration of polynucleotides.
- Volnewmer® RF — by a long way: We use PolyPhil™ as a preparatory treatment prior to Volnewmer® or as a consolidation treatment afterwards, but not during the same session (with a two-week interval).
- Stellar M22™ IPL — the following treatments can be carried out in the same session: Following IPL treatment, it acts as a regenerative layer — soothing the skin and rebuilding the skin matrix.
When using peels such as BioRePeel® or PRX-T33® in the same session, the ideal combination is: the peel first, followed by PolyPhil™.
When is PolyPhil™ not an option? +
PolyPhil™ cannot be used in the following cases:
- Active skin infections or herpes in the treatment area
- Severe fish allergy (to be assessed on a case-by-case basis)
- Autoimmune diseases or active immunotherapy
- Severe bleeding disorder or taking blood-thinning medication
- Pregnancy and breastfeeding
- Female patients under the age of 18
